To study the mechanism of phospholipid scrambling by the G proteincoupled receptor opsin


• Opsin is a highly effective phospholipid scramblase, capable of flip-flopping
lipids rapidly across a membrane bilayer (Ploier et al. Nature Commun.
7:12832. doi: 10.1038/ncomms12832).
• Extensive atomistic molecular dynamics studies suggest a ‘credit card’
mechanism by which opsin carries out lipid scrambling.
• The available postdoctoral position is for a qualified individual who will
experimentally test the credit card mechanism by targeting the pathway
suggested by the simulations. The work involves expression and
purification of opsin point mutants in HEK293 cells, reconstitution into lipid
vesicles and activity tests using an established fluorescence-based assay.
• Experience in biochemistry is essential; experience in membrane protein
purification and reconstitution is preferred.
